Light Switch Reagent PA-Cre with Magnet System

It is possible to control the function of genes in a time- and space-specific manner using light stimulation.

In the field of life sciences, there is a demand for optical manipulation techniques that achieve high temporal and spatial resolution and allow for precise control that is difficult to achieve with drug administration, as a means of functional analysis of genes and the creation of disease models. This product is a light-inducible Cre recombinase (Photoactivatable Cre; PA-Cre) that combines Cre recombinase with the light-switchable protein Magnet System. By using this product, it becomes possible to control the function of genes in a time- and space-specific manner through light stimulation.

PA-Cre with the Magnet System links the split fragments of Cre recombinase, which have lost their DNA recombination activity (split-Cre; CreN59 and CreC60), to the light-switching protein Magnet System (nMag and pMag). PA-Cre does not exhibit DNA recombination activity against loxP (base sequence) in the dark (Inactive PA-Cre). When blue light (470±20nm) is irradiated, the binding of the Magnet System (nMag and pMag) introduced into PA-Cre leads to the complementation of the split Cre recombinase (CreN59 and CreC60), allowing PA-Cre to rapidly regain its activity (Active PA-Cre). This mechanism enables the recombination reaction of DNA sequences flanked by two loxP sites to occur only in cells exposed to blue light at the time of irradiation, allowing for the time- and space-specific control of gene function.
