Hydroxyapatite chromatography is considered to be "pseudo-affinity" chromatography or "mixed-mode" ion exchange. It has been demonstrated to be an effective purification mechanism in various processes, providing biomolecular selectivity that further complements traditional ion exchange or hydrophobic interaction methods. HA Ultrogel hydroxyapatite resin is composed of cross-linked agarose beads and microcrystals of hydroxyapatite incorporated into agarose mesh. The particle size ranges from 60 to 180 μm. The agarose component of HA Ultrogel is chemically stabilized by epichlorohydrin under strongly alkaline conditions. This forms glycerol bridges between polysaccharide chains, providing the resin with excellent rigidity and stability against changes in pH, ionic strength, and high temperatures. HA Ultrogel can be regularly treated with 0.1M NaOH for regeneration and purification. The porosity of HA Ultrogel is comparable to that of agarose gel, with a size exclusion limit for globular proteins of 5,000,000 daltons. This macroporosity avoids the molecular sieving effect during separation. The resin is shipped in 1 M NaCl containing 20% ethanol and is available in a wide range of package sizes. Special packages to meet specific manufacturing requirements are also available upon request.
