Application Note: Automation of Cell-Based Assays

Drug efficacy profiling against anticancer agents using the NCI-60 cell line panel.

The NCI-60 panel is a collection of various cancer cell lines and is one of the most important tools in drug discovery for evaluating the activity of anticancer agents. While cell lines are commonly used in molecular biological assays, when combined with bioinformatics such as the NCI COMPARE database, results obtained from the NCI-60 panel can also estimate the mechanisms of drug efficacy against novel compounds. The efficacy evaluation for a large variety of compounds is initially tested at a single concentration. Hit compounds that show cell growth inhibition or cytotoxicity are then profiled in detail against multiple cell lines by creating dilution series. Tekan and the German biotech company European ScreeningPort GmbH have developed a protocol to demonstrate this cell-based assay solution. By combining expertise in cell-based assays from automated liquid dispensing to detection, they provide fast and reliable fully automated assays. After sample preparation using Fluent, cell viability was measured using luciferase detection for cellular ATP measurement (CellTiter-Glo, Promega).
