The intrinsic method for dissolution testing provides important information about the active pharmaceutical ingredient by determining the dissolution rate from a test solution with a constant pH, temperature, and surface area. The Biopharmaceutics Classification System (BCS) classifies the characteristics of a drug's solubility and membrane permeability, while the intrinsic method can characterize solubility.

The dissolution rate is expressed as the mass of the sample that dissolves per unit time in the elution test solution. The sample concentration is plotted against time and surface area as mg/min/cm², and the slope of the line becomes the intrinsic solubility constant. The intrinsic solubility constant also provides information about physical properties, crystallinity, polymorphism, solvation, and more.
