Introducing a method for the separation of stereoisomers of plasmid DNA pBR322 using anion exchange chromatography!
In general, plasmid DNA is obtained through purification from E. coli culture broth. The culture broth contains not only contaminating components such as host-derived genomic DNA, RNA, and proteins, but also open circular DNA (Open circular) and linear DNA, which are structural isomers of the target plasmid. Since these can reduce the efficiency of gene expression, clinical plasmid DNA is required to have a purity of over 90% for supercoiled DNA.
