Until now, X-ray crystallography and NMR-based analyses have primarily been used to measure protein conformation. While these analytical methods are useful, they are not applicable to all proteins and often require high-purity, large quantities of samples. Today, by effectively combining the ACQUITY M-Class HDX system with high-resolution mass spectrometry and rigorously controlling the temperature-sensitive aspects of the HDX sample preparation, we can reliably identify changes in protein conformation.
