Application Note: Compound Profiling of CYP Activity
High-density screening with the automated liquid handler Fluent.
Fluent achieves greater capacity and faster speeds. This platform offers excellent throughput and walkaway time, making it easier and more effective to conduct experiments. Tecan and Fraunhofer IME ScreeningPort have developed a protocol to demonstrate the capabilities of the Fluent laboratory automation solution for biochemical assays by combining their expertise in automated liquid handling. It has become possible to provide high-speed, reliable, and fully automated assays without the need for specialized automation personnel.
basic information
The failure of preclinical pharmaceuticals is a significant issue for pharmaceutical companies, as it accounts for a considerable portion of valuable R&D budgets. As a result, there is a need to bring compound toxicity testing into high throughput, similar to primary efficacy screening. Drug metabolism is a crucial factor in compound toxicity, and cytochrome P450 (CYP), which is predominantly expressed in hepatocytes, plays a very important role in this metabolism. Currently, there are many screening assays designed to be automated in a microplate format for profiling compounds that may induce or inhibit CYP activity. This application note describes a lead generation method using three different CYP enzymes in a 384-well microplate format and presents typical assays performed in pharmacokinetics labs. IME's ScreeningPort division is engaged in the development of cell-based biochemical assay systems, high-throughput drug discovery, and biomarker identification. Tecan has implemented automation using its unique liquid handling device, Fluent, built around the application.
