Protein analysis encompasses a variety of platforms, including mass spectrometry, ELISA, and other antibody-based approaches, as well as more recent platforms that utilize next-generation sequencing technologies. While each method offers distinct features, they differ with respect to specificity, reproducibility, multiplexing capability, and sample requirements. Olink’s Proximity Extension Assay (PEA) technology represents a distinct category of antibody-based multiplex proteomics. By combining dual antibody recognition with DNA-based signal amplification, a measurable signal is generated only when two antibodies bind in close proximity to the same target protein. This design contributes to reduced non-specific interactions while supporting high specificity. In addition, the technology enables high-sensitivity, simultaneous measurement of thousands of proteins from minimal sample volumes, making it applicable across a broad range of research phases, from discovery to validation studies.
